Immunologic Studies on the Anti-Pig Zona Antibody

Abstract
It has been considered that treatment of zonae pellucidae with anti-ovary serum containing antibody activity against zona, specific antigen(s) produce a precipitin layer on the outer surface of the zona which inhibits both sperm attachment and penetration. The presence of this precipitin layer is believed to be responsible for observed inhibition of fertilization in vitro and in vivo following either antiserum treatment of eggs or passive immunization.
With prepared rabbit anti-pig ovary and egg serum, author studied by gel diffusion, immunoelectrophoresis and indirect immunofluorescence to determine (1) antigens and cross-reactivity of the pig ovary, egg, other pig tissues and rabbit ovary, (2) the presence of specific antigen(s) in pig ovary and egg, (3) the antigenecity and presence of zona specific antigen, (4) cross-reactivity between pig zona and rabbit zona.
The results were summarized as follows:
1. Anti-pig ovary and egg serum contained antibodies cross-reacted with many pig tissue components but rabbit ovary. Pig follicular fluid and plasma were antigenically very similar with pig ovary and egg by gel diffusion and immunoelctrophoresis.
2. The presence of single specific antigen in pig ovary and egg was detected by gel diffusion and immunoelectrophoresis.
3. Maximum titer of anti-pig zona IgG antibody in anti-pig ovary and egg serum was 1:2,560 and 1:5,120, respectively. Also observed strong antigenecity of pig zona after immunization of pig eggs without adjuvant such as titer of 1:640.
4. The presence of zona specific antigen in pig was observed by indirect immunofluorescence.
5. Follicular fluid absorbed anti-pig ovary and egg serum cross-reacted with rabbit zona at a dilution of 1:80 and 1:320, respectively.
6. After passive immunization with follicular fluid absorbed anti-pig egg serum, anti-pig zona IgG antibodies were precipitated on the surface of rabbit zona.
